Burnage, who were also promoted last season, from North West One, are Saturday's opponents.
The visitors sit nine points behind York after recording just two wins from their first nine games.
On paper, York have a number of winnable games between now and Christmas and will be looking to consolidate a top half league position after the first half of the season.
York have made four changes to their squad with Tom Woffendin, Tom Hicks, Will Norris and Willem Enslin coming in to the fold.
York’s second team will be looking to back up last week's strong win over Sheffield Tigers with another good performance at home to Otley.

Selby have a home fixture against Goole in Yorkshire One.
In Yorkshire Four North/West, York Railway Institute entertain Nestle Rowntree in a North Yorkshire derby.
